Centrifugation is a process used for liquid/liquid or liquid/solid separation. Kemtrak in-line photometers can be placed at a number of locations on a centrifuge in order to:

  • increase product yield / reduce product loss
  • provide a more consistent product quality
  • increased separation efficiency

The feed stream of a centrifuge will often vary in solids content, adversely effecting the performance. By monitoring changes in the feed concentration using a Kemtrak TC007 LED turbidimeter, overloads can be prevented, bowl speed can be optimized and flow can be controlled based on solids content.

The centrate is a common location used to control and optimize a centrifugal separator and to protect downstream filtration equipment.

The concentrate may also be monitored in order to evaluate the centrate carryover or solids content.

The Kemtrak DCP007 LED photometer is an advanced dual wavelength photometer designed to accurately measure the concentration or color of light absorbing substances from 250 to 1050 nm (UV-VIS-NIR). The Kemtrak TC007 LED turbidimeter uses a long life NIR LED light source to precisely measure the optical density of turbid liquids for applications such as monitoring the solids concentration in the concentrate.

Optic fibers are utilized to pipe the light from within the analyzer enclosure to the sampling point and back. Kemtrak measurement cells are available in a range of materials and design, including DIN and sanitary TriClamp flanges.

Chromatography involves passing a mixture of substances dissolved in a mobile phase through a stationary phase, in order to separate the substances of interest. Chromatography requires accurate monitoring and control and repeatability is essential.

The Kemtrak DCP007 UV LED photometer uses a long life high performance LED light source with robust fiber optics to provide drift and noise-free measurement with very high precision.

A proprietary dual wavelength four channel measurement technique and advanced digital electronics design allows deep absorbance measurement to 5 AU. A range of shorter optical pathlengths allow for deeper absorbance measurements. Automatic compensation for sample turbidity and/or fouling of the optical windows ensures trouble free operation.

Measurement cells are available in intrinsically safe hygienic TriClamp flanges to custom manifolds with less than 1 µL internal volume for fast and accurate concentration measurement of proteins.

In-line ISO7027 turbidity measurement and control - Kemtrak DCP007 industrial process fiber optic turbidimeter

 

The Kemtrak TC007 LED Turbidimeter uses high performance long life NIR LED lamps to accurately measure the turbidity of the sample in accordance with the International Standard ISO7027 defining turbidity measurement.

As with all Kemtrak photometers, the Kemtrak TC007 is easy to install and requires no maintenance. Optic fibers are utilized to pipe the light from within the analyzer enclosure to the sampling point and back. Kemtrak flow-cell manifolds are available in a range of materials and design, including DIN and sanitary TriClamp flanges. Process probes for use in tanks are also available.


If two phases have any difference in either turbidity or absorbance between 250nm and 2500nm, then they can be separated by a Kemtrak process photometer. By monitoring either the absorbance or turbidity at strategic points in the process, immediate detection of product-product or product-water interfaces can be achieved.

Proteins

Proteins are large biological molecules comprised of one or multiple amino acids. Proteins in solution absorb ultraviolet light with an absorbance maximum at 280nm. The primary reason for this is due to the aromatic amino acids, e.g. phenylalanine, tryptophan, tyrosine and histidine.

For post chromatography column detection, UV absorption is used to detect the onset and end of proteins as they elute the column, in addition to the total volume eluted from the column. Liquid buffer and solvent gradients in the mobile phase are utilized in order to elute the sample components from the chromatography column. UV absorption is the preferred technique as the mobile phase often has no absorption at 280nm allowing a precise control of the chromatographic separation irrespective of the continuous phase composition.

It is also possible to measure the purity of biomolecules by ratio of absorbance at 280nm and 260nm. The A260/A280 ratio provides an indication of protein contamination in nucleic acid isolates and less commonly, nucleic acid contamination in protein isolates.

A Kemtrak DCP007-UV LED photomter is the recommended instrument to accurately measure protein concentration. Traditional UV photometers use powerful mercury vapor UV lamps that subject the sample to broad spectrum UV radiation and heat generated by the lamp. The resulting light that has passed through the sample then passes through a UV filter and is detected by a photodiode. The hot broad-spectrum UV radiation used on these traditional photometers destroys the sample being analyzed resulting in unknown and potentially hazardous breakdown products.


The Kemtrak DCP007-UV LED photometer uses a state of the art high performance UV LED light source and robust fiber optics. The sample is subjected to ultra-low power cold light of the exact wavelength required for the analysis, thousands of times less powerful than that used on traditional photometers. The Kemtrak UV LED light source is also modulated and can be configured to measure during very short pulses, further reducing the exposure of the sample to harmful UV radiation.

The measurement of water in alcohols or hydrocarbon solvents is an important measurement in many pharmaceutical processes. The use of near-infrared (NIR) spectroscopy for water analysis is common as NIR offers high sensitivity and measurements can be undertaken in-line providing immediate results.

Water measurements can be undertaken on hydrocarbon solvents with ppm accuracy. Proven applications include acetone, isopropyl benzene, propanol, tetrahydrofuran, and methyl-isobutyl ketone, to name just a few. Additionally, water can be measured in alcohols, such as methanol and ethanol, from trace amount to virtually 100% water.

Water has two regions of activity in the NIR region between 1000 - 2000nm. At 1400nm the O-H stretch in the water molecule is measured, while at 1900nm water has a unique peak due to a combination of O-H stretch and
H-O-H bending. The water peak at 1900 is about five times as large as the water peak at 1400.
